清宫长春丹胶囊对肾阳虚小鼠应激能力的影响(英文)

背景:依据清宫秘方研制而成的纯中药制剂清宫长春丹胶囊,具有补肾益精、强筋壮骨、延缓衰老之功效,临床用于治疗身衰体弱、精力不足、健忘易倦、头晕耳鸣、腰痛膝酸等病症。目的:观察清宫长春丹胶囊对以肌肉注射氢化可的松制备的“阳虚”模型小鼠应激能力的影响。设计:完全随机对照动物实验。地点:承德医学院中药研究所(河北省中药研究与开发重点实验室)。材料:二级昆明种小鼠160只,,体质量19~21g,雌雄各半。方法:实验于2002-02/2002-12在承德医学院中药研究所药理毒理研究室完成。采用昆明种小鼠160只,雌雄各半,随机分为游泳耐力实验组(n=60)、常压耐缺氧实验组(n=50)、单核巨噬细胞吞噬功能组(n=50)3组,每组再平均分为长春丹0.25,0.5,1.0g/kg,模型对照组及正常小鼠对照组共5个亚组。禁食12h,以肌肉注射氢化可的松25mg/kg连续7d制备“阳虚”模型,按分组给不同剂量长春丹,给药容积20mL/kg,连续给药7d,模型对照组给予等容积生理盐水灌胃,最后一次给药40min后,分别进行小鼠游泳耐力实验、小鼠常压耐缺氧时间实验、小鼠单核巨噬细胞吞噬功能的实验(尾静脉注射印度墨汁0.lmL/kg,分别于注射后2min,10min从眼眶静脉丛取血20μL,计算单核巨噬细胞炭粒廓清指数K值),观察不同剂量的长春丹对小鼠游泳时间、耐缺氧时间和单核巨噬细胞吞噬功能的影响。结果:纳入实验的160只小鼠全部进入结果统计分析,无缺失值。①长春丹胶囊0.25,0.5,1.0g/kg剂量组及正常对照组小鼠游泳时间均明显长于模型对照组[(10.94±3.79),(13.68±5.62),(14.58±5.49),(16.12±2.35),(6.45±4.87)min;t=2.522,3.367,3.586,3.979,P<0.05~0.001]。②长春丹胶囊0.25,0.5,1.0g/kg剂量组及正常对照组小鼠常压耐缺氧时间均明显长于模型对照组[(19.45±4.63),(21.18±4.25),(22.58±5.21),(23.12±4.78),(12.35±4.89)min;t=1.566,2.104,2.004,2.303,P<0.05~0.01]。③长春丹胶囊0.25,0.5,1.0g/kg剂量组及正常对照组小鼠单核巨噬细胞吞噬功能炭粒廓清指数(K)均明显高于模型对照组(0.0236±0.0106,0.0291±0.0092,0.0308±0.0086,0.0318±0.0101,0.0125±0.0081;t=2.63,4.282,4.898,4.714,P<0.05~0.01)。结论:清宫长春丹胶囊灌胃小鼠游泳时间明显延长,且可剂量依赖的延长小鼠常压耐缺氧时间,提高小鼠单核巨噬细胞的吞噬功能,显著的增强“阳虚”小鼠的应激能力。 Background: Purified Changchundan capsule, a pure Chinese medicine preparation developed according to the secret formula of Qing Palace, has the effects of nourishing the kidney, strengthening essence, strengthening bones, and delaying aging. Design: Completely randomized controlled animal experiments. Venue: Institute of Traditional Chinese Medicine, Chengde Medical College (Hebei Provincial Key Laboratory of Chinese Medicine Research and Development). MATERIALS: A total of 160 Kunming mice of the second grade were used, with body weight of 19-21 g, half male and half female. METHODS: The experiment was performed at the Institute of Pharmacology and Toxicology, Institute of Traditional Chinese Medicine, Chengde Medical College from 2002-02 to 2002-12. 160 Kunming mice were used, male and female, and randomly divided into swimming endurance test group (n=60), normoxic hypoxia test group (n=50), mononuclear macrophage phagocytosis group (n=50). 3 groups, each group was evenly divided into 0.25, 0.5, and 1.0 g/kg of Changchun Dan, and the model control group and the normal mouse control group consisted of 5 subgroups. After fasting for 12 hours, the “Yang-Deficiency” model was prepared by intramuscular injection of hydrocortisone 25 mg/kg for 7 days. According to different doses of vinpoceine, the dose volume was 20 mL/kg, continuous administration for 7 days. The model control group was given equal volume physiology. Saline was intragastrically administered. After 40 minutes of the last administration, mice were subjected to swimming endurance test, mouse hypobaric hypoxia tolerance test, and mononuclear macrophage phagocytic function test (Tail vein injection of Indian ink 0.1 mL/well.
