目的探讨has-miR-429、has-miR-127-3 p及has-miR127-5 p在不同宫颈癌细胞中的表达水平。方法用SiHa、Hela和C33A三种宫颈癌细胞株,提取和纯化mRNA,逆转录成cDNA,在测定最佳反应条件后,采用实时荧光定量PCR技术测定has-miR-429、has-miR-127-3p及has-miR127-5p的表达情况,结合内参U6进行相对定量分析。结果 miR-429、miR-127-3p及miR127-5p在SiHa细胞相对于Hela细胞表达为(0.0031±0.0002),(59.3786±3.7024)和(0.0524±0.0148),相对于C33A细胞表达为(0.0059±0.0005),(15.5625±1.2780)和(0.0334±0.0103)。结论 has-miR-429、has-miR-127-3p及has-miR127-5p可能与宫颈癌的HPV感染相关。
Objective To investigate the expression of has-miR-429, has-miR-127-3 p and has-miR127-5 p in different cervical cancer cells. Methods The mRNAs of hsa-miR-429 and has-miR-127 were determined by real-time fluorescence quantitative PCR after the optimal reaction conditions were extracted and purified from the three cervical cancer cell lines with SiHa, Hela and C33A. -3p and has-miR127-5p expression, combined with the reference U6 relative quantitative analysis. Results The expression of miR-429, miR-127-3p and miR127-5p were (0.0031 ± 0.0002), (59.3786 ± 3.7024) and (0.0524 ± 0.0148) in SiHa cells compared with Hela cells and (0.0059 ± 0.0005), (15.5625 ± 1.2780) and (0.0334 ± 0.0103). Conclusion The expression of has-miR-429, has-miR-127-3p and has-miR127-5p may be related to HPV infection of cervical cancer.
