目的通过免疫组织化学染色的方法了解Thy1在肺癌组织中的表达状况,分析其对于非小细胞肺癌患者预后的影响,并了解Thy1与p53表达之间的相关性,为Thy1在肺癌中表达机理的进一步研究提供思路。方法以组织芯片的形式,采用免疫组化信号催化放大法,对175例包括不同病理类型的肺组织标本中Thy1的表达情况进行检测,并对其中的91例非小细胞肺癌进行Thy1和p53的联合检测。分析这两种蛋白表达之间的相关性,结合临床病理资料及随访结果分析Thy1与患者生存预后的关系。结果Thy1抗体特异性地在不同病理类型肺癌细胞的胞核中有强阳性染色,89例(56.7%)为Thy1细胞核染色阳性。91例非小细胞肺癌患者预后的单因素和多因素分析显示,TNM分期、淋巴结转移和Thy1在细胞核中高表达是影响患者预后的独立因素。分层研究提示,无淋巴结转移组中,Thy1细胞核阴性患者的生存时间明显长于阳性患者(分别为46.42个月和38.56个月,P=0.0207)。Thy1和p53两者表达之间存在显著的相关性(P<0.001)。结论Thy1在肺癌细胞核内特异性高表达是非小细胞肺癌患者预后的影响因素之一。Thy1在细胞核内的异常高表达可以作为肺癌病理诊断的标记物。
Objective To investigate the expression of Thy1 in lung cancer by immunohistochemical staining and analyze the influence of Thy1 on the prognosis of patients with non-small cell lung cancer and to understand the correlation between Thy1 and p53 expression and to investigate the mechanism of Thy1 expression in lung cancer Further research to provide ideas. Methods Tissue microarrays were used to detect the expression of Thy1 in 175 cases of lung tissue specimens with different pathological types by immunohistochemical signal amplification. Thy1 and p53 were detected in 91 non-small cell lung cancer Joint testing. To analyze the correlation between the expression of these two proteins and to analyze the relationship between Thy1 and the prognosis of patients with clinicopathological data and follow-up results. Results Thy1 antibody specifically in the different pathological types of lung cancer cells in the nucleus strong positive staining, 89 cases (56.7%) were positive for Thy1 nuclear staining. The prognosis of 91 patients with non-small cell lung cancer by single factor and multivariate analysis showed that TNM staging, lymph node metastasis and high expression of Thy1 in the nucleus were independent prognostic factors. Hierarchical studies suggest that the survival of patients with negative Thy1 nucleotides was significantly longer than that of positive patients without lymph node metastasis (46.42 months and 38.56 months, respectively, P = 0.0207). There was a significant correlation between Thy1 and p53 expression (P <0.001). Conclusion The high expression of Thy1 in the nucleus of lung cancer cells is one of the prognostic factors in patients with non-small cell lung cancer. Thy1 abnormally high expression in the nucleus can be used as a marker for pathological diagnosis of lung cancer.
