[目的]比较朝鲜族与汉族丙型肝炎病毒RNA(HCV-RNA)阳性者白细胞介素(IL)28BSNP rs12979860的表型分布,探讨延边朝鲜族易感HCV的可能机制.[方法]选择HCV-RNA阳性的朝鲜族98例、汉族92例及检查证实为肝炎病毒指标阴性的健康朝鲜族53例与汉族50例列入研究对象.应用焦磷酸测序法对IL-28B基因多态性SNP rs12979860位点进行检测,分析朝鲜族与汉族HCV-RNA阳性者及健康人群的C等位基因频率和基因型比例,并对基因型与HCV亚型、HCV-RNA定量、肝功能及血象间的相关性进行分析.[结果]朝鲜族HCV-RNA阳性者中CC型89例、TC+TT型9例,汉族HCV-RNA阳性者中CC型83例、TC+TT型9例;健康人群中CC型92例、TC型11例;朝鲜族与汉族HCV-RNA阳性者及健康组基因频率间无明显差异,基因型与HCV亚型、HCV-RNA定量、肝脏生物化学指标间差异亦均无统计学意义(P>0.05).[结论]延边地区朝鲜族与汉族IL-28BSNP rs12979860位点主要表现为CC型,提示IL-28BSNP rs12979860位点C等位基因频率为优势基因表型,但它不能解释朝鲜族易感HCV的原因.
[Objective] To investigate the phenotype distribution of interleukin (IL) 28BSNP rs12979860 in Korean and Han patients with hepatitis C virus (HCV) RNA and to explore the possible mechanism of susceptibility HCV in Yanbian Korean. [Methods] HCV- 98 cases of Korean Han, 92 cases of Han and check to be negative for hepatitis virus, healthy Korean 53 and Han 50 cases were included in the study.Using pyrosequencing of IL-28B gene polymorphism SNP rs12979860 Point detection and analysis of C allele frequencies and genotypes in HCV-RNA positive and healthy people of Korean and Han nationality, and the correlation between genotype and HCV subtypes, HCV-RNA quantitation, liver function and blood picture .Among them, 89 cases were CC genotype and 9 cases were TC + TT genotype in Korean HCV-RNA positive group, 83 cases were CC genotype and 9 cases were TC + TT genotype in Han nationality with HCV-RNA positive. In CC genotype 92 cases, TC type 11 cases; Korean and Han HCV-RNA positive and healthy group no significant difference in gene frequency, genotype and HCV subtypes, HCV-RNA quantitative, liver biochemical indicators were also no significant difference (P> 0.05). [Conclusion] The rs12979860 locus of IL-28BSNP in Korean and Han nationality in Yanbian area is mainly Now, the CC genotype, suggesting that IL-28BSNP rs12979860 C allele frequency of the dominant phenotype, but it can not explain why Korean susceptible to HCV.
