目的探讨坎地沙坦对动脉粥样硬化(AS)大鼠血脂以及血管基质金属蛋白酶2(MMP-2)、血管内皮生长因子(VEGF)表达的影响。方法饲喂高脂饲料建立AS大鼠模型。随机分为正常组(A组)、模型组(B组)、坎地沙坦低(1 mg.kg-1.d-1,C组)、中(5 mg.kg-1.d-1,D组)、高(10 mg.kg-1.d-1,E组)剂量组。12周后腹主静脉取血检测各组血脂;取腹主动脉标本采用免疫组化方法测定大鼠血管MMP-2和VEGF的表达。结果与B组相比,C、D、E组血脂(胆固醇、甘油三酯、低密度脂蛋白)均降低(P<0.05,P<0.01),但无剂量依赖性;D、E组血管MMP-2、VEGF的表达减少(P<0.05)。结论坎地沙坦可降低AS大鼠的血脂和血管MMP-2、VEGF的表达。
Objective To investigate the effects of candesartan on the blood lipids and the expression of matrix metalloproteinase 2 (MMP-2) and vascular endothelial growth factor (VEGF) in atherosclerotic rats. Methods High fat diet was used to establish AS rat model. The rats were randomly divided into normal group (group A), model group (group B), candesartan low (1 mg.kg-1.d-1, group C) , D group), high (10 mg.kg-1.d-1, E group) dose group. After 12 weeks, blood samples were taken from abdominal aorta and blood was collected to measure blood lipids. The abdominal aorta specimens were used to detect the expression of MMP-2 and VEGF by immunohistochemistry. Results Compared with group B, the levels of lipids (cholesterol, triglyceride and low density lipoprotein) in groups C, D and E were decreased (P <0.05, P <0.01) -2, VEGF decreased (P <0.05). Conclusion Candesartan can reduce the blood lipids and vascular MMP-2, VEGF expression in AS rats.
