目的建立快速高分离度液相色谱(RRLC)同时测定肚痛健胃整肠丸中甘草苷、橙皮苷和甘草酸含量的方法。方法采用BDS HYPERSIL C1(84.6 mm×100 mm,2.4μm)色谱柱;流动相为乙腈-0.2%磷酸,梯度洗脱;流速为1.0 mL.min-1;检测波长为0~9 min,276 nm;9~14 min,250 nm;柱温为25℃;进样体积为4.0μL,检测器响应时间1 s。结果样品中甘草苷、橙皮苷和甘草酸在进样量分别为0.0112~0.2800μg、0.0435~1.0875μg、0.0630~1.5750μg范围内有良好的线性关系(r=0.9999)。平均回收率分别为99.1%,99.66%,99.26%(n=6);RSD分别为0.79%,1.61%,0.99%。结论本方法可在较短的时间内快速得到准确的结果,有效控制肚痛健胃整肠丸的产品质量。
Objective To establish a method for the simultaneous determination of liquiritin, hesperidin and glycyrrhizin in abdominal pain, stomach and intestine with pellets by fast and high resolution liquid chromatography (RRLC). Methods The chromatographic column was BDS HYPERSIL C1 (84.6 mm × 100 mm, 2.4 μm). The mobile phase consisted of acetonitrile-0.2% phosphoric acid with a gradient of 1.0 mL.min-1. The detection wavelength was 0-9 min and the wavelength was 276 nm ; 9-14 min, 250 nm; column temperature was 25 ° C; injection volume 4.0 μL, detector response time 1 s. Results There was a good linearity (r = 0.9999) for the contents of glycyrrhizin, hesperidin and glycyrrhizic acid in the range of 0.0112 ~ 0.2800μg, 0.0435 ~ 1.0875μg and 0.0630 ~ 1.5750μg respectively. The average recoveries were 99.1%, 99.66% and 99.26%, respectively (n = 6). The RSDs were 0.79%, 1.61% and 0.99%, respectively. Conclusion This method can quickly and accurately obtain the result in a short period of time and effectively control the product quality of abdominal pain, stomach and intestine.
