目的研究跨损伤DNA合成通路中的关键基因RAD6、RAD18、PCNA和UBC13在人食管鳞癌与癌旁组织中的表达差异。方法采用SYBRGreen实时定量PCR技术检测60例食管鳞癌患者癌组织和其中的24例癌旁组织中RAD6、RAD18、PCNA和UBC13基因表达情况。结果 RAD6和UBC13在食管鳞癌与癌旁组织中的表达差异均无统计学意义(均P>0.05);RAD18和PCNA的表达在食管鳞癌中均显著上升,与癌旁组织相比差异均有高度统计学意义(均P<0.01)。结论 RAD18和PCNA的表达升高可能是食管鳞癌的一个特征。RAD18和PCNA表达的升高可能与食管鳞癌中跨损伤DNA合成途径作用增强有关。
Objective To study the expression differences of the key genes RAD6, RAD18, PCNA and UBC13 in human esophageal squamous cell carcinoma and paracancerous tissues. Methods SYBR Green real-time PCR was used to detect the expression of RAD6, RAD18, PCNA and UBC13 in 60 cases of esophageal squamous cell carcinoma and 24 cases of paracancerous tissues. Results There was no significant difference in the expression of RAD6 and UBC13 between esophageal squamous cell carcinoma and adjacent normal tissues (all P> 0.05). The expressions of RAD18 and PCNA in esophageal squamous cell carcinoma were significantly increased There was a high degree of statistical significance (all P <0.01). Conclusion The increased expression of RAD18 and PCNA may be a characteristic of esophageal squamous cell carcinoma. The increased expression of RAD18 and PCNA may be related to the enhancement of DNA synthesis pathway in trans-injury of esophageal squamous cell carcinoma.
