目的观察和营安心方对衰老大鼠血清和脑组织白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)的影响,探讨和营安心方对脑组织的保护作用。方法 48只Wistar雄性大鼠随机分为正常组、模型组、维生素E组及和营安心方高、中、低剂量组。除正常组外,其他组颈背部皮下注射D-半乳糖125 mg·kg~(-1)·d~(-1)制备大鼠衰老模型,各组给予相应药物灌胃,正常组和模型组给予等量生理盐水灌胃,连续8周。酶联免疫吸附法(ELISA)检测各组大鼠血清和脑组织中IL-6和TNF-α含量。结果与正常组比较,模型组血清和脑组织中IL-6和TNF-α含量增高,差异有统计学意义(P<0.05);与模型组比较,各给药组血清和脑组织中IL-6和TNF-α含量降低,差异有统计学意义(P<0.05),和营安心方高剂量组作用最为显著。结论和营安心方可下调血清和脑组织中IL-6和TNF--α的含量,通过减轻炎症反应,对脑组织起到保护的作用。
Objective To observe the effects of Yinganxinfang on serum and brain tissue interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) in aging rats and the protective effect of Yingan Xinfang on brain tissue. Methods Forty - eight Wistar male rats were randomly divided into normal group, model group, vitamin E group and HOPE high, medium and low dose groups. In addition to the normal group, other groups were injected subcutaneously with D-galactose 125 mg · kg -1 · d -1 on the back of the neck to prepare the model of aging in rats. The rats in each group were given gavage, normal and model groups Give the same amount of saline gavage for 8 weeks. The levels of IL-6 and TNF-α in serum and brain tissue of rats in each group were detected by enzyme-linked immunosorbent assay (ELISA). Results Compared with the normal group, the levels of IL-6 and TNF-α in the serum and brain tissue of the model group were significantly increased (P <0.05). Compared with the model group, the contents of IL- 6 and TNF-α, the difference was statistically significant (P <0.05), and Ying’an Xinfang high dose group had the most significant effect. Conclusions And Yinganxin Decoction can reduce the content of IL - 6 and TNF - α in serum and brain tissue and play a protective role on brain tissue by reducing the inflammatory reaction.
