目的:探讨人前列腺癌细胞转移潜能与其环氧化酶-2(COX-2)表达之间的关系,及环氧化酶-2与前列腺癌患者预后的关系。方法:用运免疫组化及反转录聚合酶链式反应(RT-PCR)检测两种不同转移潜能人前列腺癌细胞PC3及PC3M中COX-2mRNA及其蛋白的表达以及在肿瘤坏死因子-α(TNF-α)刺激下两者的表达是否有所变化。结果:PC3细胞在无TNF-α刺激时,检测不到COX-2mRNA的表达,其蛋白表达也为阴性,而在TNF-α刺激4h后,COX-2mRNA及其蛋白的表达均为阳性。而在PC3M细胞中无TNF-α刺激时,COX-2mRNA及其蛋白的表达已为阳性,在TNF-α刺激4h后,COX-2mRNA及其蛋白的表达均有增强。结论:COX-2可能在前列腺癌的发生发展中起着重要的作用,抑制COX-2的表达,或许能够降低前列腺癌的恶性程度甚至能够阻止其发生。这对前列腺癌患者生存期内创造良好的生活质量及前列腺癌的防治提供有力的理论依据。
Objective: To investigate the relationship between the metastatic potential of human prostate cancer cells and the expression of cyclooxygenase-2 (COX-2) and the relationship between cyclooxygenase-2 and the prognosis of patients with prostate cancer. Methods: The expression of COX-2 mRNA and protein in prostate cancer cell lines PC3 and PC3M with different metastatic potentials were detected by immunohistochemistry and reverse transcriptase-polymerase chain reaction (RT-PCR) (TNF-a) stimulated the expression of the two have changed. Results: The expression of COX-2 mRNA and protein was negative in PC3 cells without TNF-α stimulation. However, the expression of COX-2 mRNA and protein was positive in PC3 cells stimulated with TNF-α for 4 h. However, the expression of COX-2 mRNA and protein was positive in the absence of TNF-α stimulation in PC3M cells, and the expression of COX-2 mRNA and protein was enhanced 4h after TNF-α stimulation. CONCLUSION: COX-2 may play an important role in the development of prostate cancer. Inhibition of COX-2 expression may reduce the malignancy of prostate cancer and even prevent its occurrence. This provides a good theoretical basis for creating a good quality of life and prevention and treatment of prostate cancer during the survival of patients with prostate cancer.
