应用短时交配法建立大鼠妊娠模型的效果评价

目的探讨短时交配法建立孕鼠模型的可行性,为体内发育毒性研究提供基础。方法将120只雌鼠与40只雄鼠分批次按照雌雄2∶1的比例合笼,合笼时间为早上9:00,2 h后用阴道涂片法检查雌鼠的受孕情况,以观察到精子为判断标准。记录每次合笼动物数、受孕动物数,对孕鼠单笼饲养,每3 d称1次体重及摄食量,于孕21 d(Gestation Day,GD21)处死孕鼠,并观察记录动物的实际受孕情况及仔鼠情况。结果雌鼠单次交配平均受孕率为14.1%,饲养14 d后进入受孕高峰期,40 d后受孕率明显降低。孕鼠体重、摄食量的变化符合胚胎发育的各阶段需求,胎鼠发育情况良好未见畸形发生。结论短时交配法具有可行性,所获孕鼠及分娩胎鼠的生长情况良好,可以满足发育毒性试验的需求,尤其对需严格要求胚胎发育时间的试验来说是一种较为理想的动物交配方式。 Objective To investigate the feasibility of establishing a pregnant rat model by short-term mating method and provide a basis for the study of developmental toxicity in vivo. Methods 120 female rats and 40 male rats were caged in the ratio of male and female 2: 1 in batches. The cages were caged at 9:00 am and examined by vaginal smear. Sperm as a criterion. Results The average pregnancy rate of single mating in female rats was 14.1%, and reached the peak of conception after 14 days of feeding. The pregnancy rate of female rats decreased significantly after 40 days. Pregnant mice body weight, food intake changes in line with the needs of various stages of embryonic development, fetal development was good without deformity. Conclusion The short-time mating method is feasible. The growth of pregnant mice and delivery fetus is good, which can meet the needs of developmental toxicity test, and is an ideal animal mating test especially for the test that requires strict time of embryo development the way.
