本文应用结肠癌、伴异型增生的结肠腺瘤及正常结肠粘膜组织的石蜡切片作间接法免疫过氧化酶染色,以测定一批肿瘤伴随抗原的抗体的肿瘤特异性。所用抗体包括:抗癌胚抗原(CEA)的多克隆抗体及单克隆抗体Mab3851、Mab27;抗蛋白质—碳水化合物的单克隆抗体MabC216、MabC242;抗脂质—碳水化合物的单克隆抗体MabC50、MabC19-9。实验结果发现三个单克隆抗体(Mab3851、Mab27及MabC216)的肿瘤特异性与正常对照有显著性差异。其增高的染色频率也见于异型增生的粘膜。而MabC242则逊於前三者。抗CEA的多克隆抗体、单克隆抗体MabC50及Mab19-9无肿瘤特异性。结果表明前三个抗体在鉴别结肠粘膜的良恶性质上,尤其在识别癌前病变上,具有一定的参考价值。
In this paper, colon cancer, colon adenoma with dysplasia and normal colon mucosal tissue were used for indirect immunoperoxidase staining to determine the tumor specificity of a group of tumor-associated antigens. The antibodies used include: anti-carcinoembryonic antigen (CEA) polyclonal antibodies and monoclonal antibodies Mab3851, Mab27; anti-protein-carbohydrate monoclonal antibodies MabC216, MabC242; anti-lipid-carbohydrate monoclonal antibodies MabC50, MabC19- 9. The experimental results showed that the tumor specificity of the three monoclonal antibodies (Mab3851, Mab27, and MabC216) was significantly different from the normal control. The increased staining frequency is also seen in dysplastic mucosa. MabC242 is inferior to the first three. Anti-CEA polyclonal antibodies, monoclonal antibodies MabC50 and Mab19-9 were tumor-specific. The results showed that the first three antibodies have certain reference value in distinguishing the benign and malignant nature of the colonic mucosa, especially in recognizing precancerous lesions.
