目的:建立氯霉素搽剂中氯霉素含量测定的方法。方法:采用反相高效液相色谱法,使用Agilent ZORBAX SB-C_(18)柱(250 mm×4.6 mm,5μm),流动相为0.01 mol·L~(-1)庚烷磺酸钠缓冲溶液(用磷酸调节pH至2.5)-甲醇(60:40),检测波长277nm。结果:氯霉素搽剂在8.4~420.0μg·ml~(-1)范围内线性关系良好(r=0.999 8,n=5),平均回收率为98.2%,RSD=1.3%。结论:本方法简便、结果准确、重复性好,适用于氯霉素搽剂中氯霉素含量的测定。
Objective: To establish a method for the determination of chloramphenicol in chloramphenicol liniment. Methods: An Agilent ZORBAX SB-C 18 column (250 mm × 4.6 mm, 5 μm) was used with reversed-phase high performance liquid chromatography. The mobile phase consisted of 0.01 mol·L -1 heptane sulfonate buffer (Adjusted to pH 2.5 with phosphoric acid) -methanol (60:40) at a detection wavelength of 277 nm. RESULTS: Chloramphenicol liniment showed good linearity (r = 0.999 8, n = 5) in the range of 8.4-420.0 μg · ml -1 with an average recovery of 98.2% and a RSD of 1.3%. Conclusion: The method is simple, accurate and reproducible. It is suitable for the determination of chloramphenicol in chloramphenicol liniment.
