目的研究表皮生长因子(EGF)在雌激素受体(ER)阳性及阴性乳腺癌细胞株中对ER表达的影响及其可能的机制。方法以逆转录-聚合酶链反应(RT-PCR)技术分别研究EGF途径以及抑制该途径的信号传导后,对乳腺癌细胞株中ERαmRNA的影响。结果在ER阳性乳腺癌细胞株中,EGF能显著抑制ERαmRNA的表达(P<0.05),而通过抑制表皮生长因子受体(EG- FR)、磷脂酰肌醇3激酶(PI3K)阻断EGF信号传导可减弱上述抑制作用(P<0.05);在ER阴性乳腺癌细胞株中,ERαmRNA无显著变化。结论EGF能够明显抑制ER阳性乳腺癌细胞株中ER的表达,这种抑制作用可能通过EGFR、蛋白激酶B(PKB,又称AKt)信号传导途径完成;这种作用在ER阴性乳腺癌细胞株中并不明显。
Objective To study the effect of epidermal growth factor (EGF) on ER expression in estrogen receptor (ER) positive and negative breast cancer cell lines and its possible mechanism. Methods The effects of EGF pathway and ERαmRNA in breast cancer cell lines were studied by reverse transcription-polymerase chain reaction (RT-PCR). Results EGF could significantly inhibit the expression of ERαmRNA in ER-positive breast cancer cell lines (P <0.05), but inhibit EGF signaling by inhibiting epidermal growth factor receptor (EGFR) and phosphatidylinositol 3-kinase (PI3K) (P <0.05). In ER-negative breast cancer cell lines, there was no significant change in ERαmRNA. Conclusion EGF can significantly inhibit the expression of ER in ER-positive breast cancer cell lines. This inhibition may be mediated by EGFR and protein kinase B (AKT) signal transduction pathway. This effect may be related to ER-negative breast cancer cell lines Not obvious.
