目的:建立同时测定酸枣仁分散片中酸枣仁皂苷A、B成分含量的HPLC-ELSD方法。方法:应用高效液相色谱法,采用Venusil XBP-C18色谱柱(4.6mm×250mm,5μm);以乙腈(A)-水(0.1%乙酸)(B)为流动相梯度洗脱;流速1.0mL.min-1,柱温25℃,蒸发光散射检测器的漂移管温度为100℃,载气流速为2.9L.min-1。结果:酸枣仁皂苷A在1.12~11.24μg.g-1(r=0.9996)、酸枣仁皂苷B在1.36~13.56μg.g-1(r=0.9997)范围内线性关系良好,平均加样回收率(n=6)分别为:101.25%、98.63%;RSD分别为1.49%、2.57%。结论:本法快速简便,精密度好,灵敏度高,在同一色谱条件下实现多指标不同成分的同时测定,为酸枣仁分散片的全面质量评价提供参考。
OBJECTIVE: To establish a HPLC-ELSD method for the simultaneous determination of jujuboside A and component B in Suanzaoren dispersible tablets. Methods: High performance liquid chromatography (HPLC) was performed on a Venusil XBP-C18 column (4.6 mm × 250 mm, 5 μm). The mobile phase was eluted with acetonitrile-water (0.1% acetic acid) .min-1, column temperature 25 ℃, evaporative light scattering detector drift tube temperature of 100 ℃, carrier gas flow rate of 2.9L.min-1. Results: Jujuboside A showed a good linearity in the range of 1.12 ~ 11.24μg.g-1 (r = 0.9996) and jujuboside B in the range of 1.36 ~ 13.56μg.g-1 (r = 0.9997) (n = 6) were 101.25% and 98.63%, respectively; RSD was 1.49% and 2.57% respectively. Conclusion: The method is rapid, simple, accurate and sensitive. Simultaneous determination of different components under the same chromatographic conditions can provide a reference for the overall quality evaluation of Spinach Radish Dispersible Tablets.
