食用菌制种容器,目前国内一般都用塑料袋、瓶及各类玻璃瓶。相比之下,玻璃容器的制种效果要好些。玻璃容器通常使用的有特制菌种瓶、输液瓶、罐头瓶等,特制菌种瓶在装料、取料接种操作上较方便,污染率较低,但价格比较贵;输液瓶在接种上与试管效果一样,污染率最小,但装料和取料时不方便,很费工时;罐头瓶价格便宜且易回收,装料、取料特别方便,但接种操作不方便,且接种污染高。针对以上问题,笔者将实践中摸索出利用罐头瓶制种且行之有效方法介绍如下: 1 准备好有丝口盖的罐头瓶,将瓶盖取下,用冲子(冲头直径2cm或根据需要而定),在盖中心冲一个孔。 2 装料时将盖拧开,料装满后将盖拧紧,并用棉花将圆孔塞住(稍紧),再用两层纸将瓶盖包封住。 3 将装好的料瓶置高压锅内灭菌,灭菌后在接种箱(室)接种,把瓶盖上面的纸取掉,拔出棉塞,过酒精灯火焰接入菌种,再塞上棉塞,不再盖纸(这样菌丝长势好),然后进行培养。当菌丝长满瓶需用时,可拧开罐
