目的 探讨脑梗死和偏头痛患者血小板内钙浓度及其凝血酶刺激后钙浓度的变化。方法 应用Fura 2负载血小板双波长扫描 ,连续测定了 2 0例脑梗死患者与 2 0例偏头痛患者血小板静息态胞浆游离钙浓度 ([Ca2 + i])及观察凝血酶诱导后血小板 [Ca2 + i]动态变化 ,并记录了凝血酶刺激后血小板 [Ca2 + i]上升时间 (t)。结果 脑梗死患者和偏头痛患者血小板静息态、凝血酶诱导后 [Ca2 + i]及凝血酶诱导后较静息态的[Ca2 + i]净增量△ [Ca2 + i]均显著高于各自的对照组。脑梗死患者和偏头痛患者比较 ,上述指标差异不显著 ;脑梗死组凝血酶刺激后血小板 [Ca2 + i]上升时间较对照组明显延长 ,而偏头痛组较对照组明显缩短 ,脑梗死组较偏头痛组延长。结论 血小板 [Ca2 + i]异常增高可能是该两种疾病血小板异常的共同机制之一 ,血小板受刺激后 [Ca2 + i]上升时间的差异则可能反应了这两种疾病的本质区别
Objective To investigate the changes of intra-platelet calcium concentration and thrombin-stimulated calcium concentration in patients with cerebral infarction and migraine. Methods Fura 2 loaded platelet dual wavelength scanning was used to measure the platelet resting free cytoplasm free calcium concentration ([Ca2 + i]) in 20 patients with cerebral infarction and 20 migraine patients, and to observe the effect of thrombin-induced platelet [ Ca2 + i] dynamic changes, and recorded after thrombin stimulation platelet [Ca2 + i] rise time (t). Results The resting state of platelets, [Ca2 + i] after thrombin induction and the [Ca2 + i] net increase △ [Ca2 + i] after thrombin induction were significantly higher in patients with cerebral infarction and migraine The respective control group. Compared with the control group, the rise time of platelet [Ca2 + i] in thrombin-induced cerebral infarction group was significantly longer than that in the control group, while the migraine group was shorter than the control group obviously Migraine group extended. Conclusion The abnormal increase of platelet [Ca2 + i] may be one of the common mechanisms of platelet abnormalities in these two diseases. The difference in the rise of [Ca2 +] after platelet stimulation may reflect the essential difference between the two diseases
