目的观察孟根乌森乌日乐对咪喹莫特诱导的银屑病样皮损的治疗作用及其机制。方法 BALB/c雌性小鼠50只,随机分为正常对照组、模型组、低、中、高3个剂量组,每组各10只。模型组及低、中、高3个剂量组小鼠背部涂抹咪喹莫特乳膏21 mg,每天1次,连续7 d;正常对照组小鼠背部涂抹凡士林21mg,每天1次,连续7 d。正常对照组予以0.3%羧甲基纤维素钠2 m L/100 g,每天一次,灌胃给药;模型组予以0.3%羧甲基纤维素钠2 m L/100 g,每天一次,灌胃给药;低、中、高3个剂量组分别予以0.33,0.66,1.32 g·kg-1孟根乌森乌日乐,每天一次,灌胃给药。3组均连续给药21 d。用免疫组织化学法检测皮损组织的增殖细胞核抗原、转化生长因子-α、肿瘤坏死因子-α、血小板内皮细胞黏附分子、血管内皮生长因子、Toll样受体2。结果低、中、高3个剂量组的6种免疫组织因子均高于模型组,且高剂量的效果好于中剂量,中剂量的效果好于低剂量。结论孟根乌森乌日乐对咪喹莫特诱导小鼠银屑病样皮损有较好的治疗作用。
Objective To observe the therapeutic effect and mechanism of Meganwu Wulile on imiquimod-induced psoriasis-like lesions. Methods Fifty BALB / c female mice were randomly divided into normal control group, model group, low dose, middle dose and high dose groups, 10 in each group. Mice in model group and low, medium and high dose groups were given imiquimod cream 21 mg once daily for 7 days. In the control group, 21 mg of petrolatum was applied to the back of the mice once a day for 7 days . The normal control group was given 0.3% sodium carboxymethyl cellulose 2 m L / 100 g, once a day, intragastric administration; model group was given 0.3% sodium carboxymethyl cellulose 2 m L / 100 g, once daily, Administration; low, middle and high dose groups were given 0.33,0.66,1.32 g · kg-1 Menggen Ursolum daily, once a day, intragastric administration. The three groups were given 21 consecutive days. Proliferating cell nuclear antigen, transforming growth factor-α, tumor necrosis factor-α, platelet endothelial cell adhesion molecule, vascular endothelial growth factor and Toll-like receptor 2 were detected by immunohistochemistry. Results The results showed that 6 kinds of immunohistochemical factors in low, medium and high dose groups were higher than those in model group, and the effect of high dose was better than that of middle dose. The effect of medium dose was better than that of low dose. Conclusion Menggen Wusen Würmel has a good therapeutic effect on imiquimod-induced skin lesions in mice with psoriasis.
