从长白山白眉蝮蛇蛇毒中纯化具有溶栓功效的纤溶酶。方法:用 DEAE Sephadex A50离子交换、Sephadex G75凝胶过滤层析和 DEAE Sephadex A 50三步柱层析分离方法,对长白山白眉蝮蛇蛇毒进行分离纯化,得到了一种纤溶酶活性组分。结果:经SDS-聚丙烯酰胺凝胶电泳和基质辅助激光解吸电离飞行时间质谱表征该酶为单一蛋白,其分子量为 23.367 kD。结论:为进一步研究其结构和功能提供了可靠依据。
Purification of fibrinolytic enzymes with thrombolytic activity from Changbaishan Eriophora venom. METHODS: Separation and purification of venom of Changbai Mountain were carried out using DEAE Sephadex A50 ion exchange, Sephadex G75 gel filtration chromatography and DEAE Sephadex A 50 three-step column chromatography. A plasmin activity fraction was obtained. RESULTS: The SDS-polyacrylamide gel electrophoresis and matrix-assisted laser desorption ionization time-of-flight mass spectrometry characterized the enzyme as a single protein with a molecular weight of 23.367 kD. Conclusion: To provide a reliable basis for further study of its structure and function.
