5-氟尿嘧啶(5-Fu)为临床常用的抗代谢类抗肿瘤药物,能影响DNA的合成,有较强的广谱抗癌作用,是目前临床上常用的抗癌药物之一。但5-Fu有较强的毒副作用,如胃肠道反应,骨髓抑制,严重时全身血象下降,少数可能有小脑变性,共济失调等,而且5-Fu排泄过快,使其长期应用受到限制。为了延长药效,降低毒副作用,国内外学者进行了大量的研究工作。制成5-Fu肝素钠-白蛋白毫微粒(nanoparticles,NP),是降低毒性,提高疗效的有效方法之一,有人对5-Fu白蛋白微球做了大量工作,动物实验表明,其具有肝和肺靶向性。
5-Fluorouracil (5-Fu) is a commonly used anti-metabolic antitumor drug in clinic, which can affect the DNA synthesis and has a broad spectrum of anti-cancer effects. It is one of the commonly used anti-cancer drugs in clinical practice. But 5-Fu has strong side effects, such as gastrointestinal reactions, bone marrow suppression, systemic blood loss in severe cases, a few may have cerebellar degeneration, ataxia, etc., and 5-Fu excretion too fast, so its long-term use by limit. In order to prolong the efficacy and reduce the side effects, scholars at home and abroad have carried out a large amount of research work. 5-Fu heparin sodium-albumin nanoparticles (nanoparticles, NP) is one of the effective methods to reduce toxicity and improve curative effect. Some people have done a lot of work on 5-Fu albumin microspheres. Animal experiments show that it has Liver and lung targeting.
