探讨独活及其醇提物延缓脑老化的自由基和免疫炎症方面的作用机制 ,为延缓机体衰老提供实验依据。10月龄昆明种小鼠随机分为 5组 ,采用D 半乳糖脑老化模型 ,给药 8周 ,水迷宫试验测定学习记忆能力 ,定磷法与薄层层析法测定大脑皮层、海马、纹状体的生物膜磷脂组分 ,放免法测定血清IL 2含量。结果显示 ,各组小鼠造模后水迷宫实验错误次数明显增加 (P <0 0 1) ,独活药物组的迷宫测试结果均明显低于模型对照组 (P <0 0 1) ;在大脑皮层、海马、纹状体部位 ,模型组小鼠PC含量明显降低 ,SM含量增加 ,膜老化指数SM /PC明显增高 ,用药后 ,PC含量增加 ,SM含量降低 ,SM /PC降低 ;模型对照组小鼠IL 2含量明显低于正常对照组 (P <0 0 1) ,独活药物组的结果均明显高于模型对照组 (P <0 0 1) ,独活醇提物组与正常对照组无差别 (P >0 0 5 )。提示 :独活及其醇提物通过修复大脑皮层、海马、纹状体不同部位的膜磷脂结构 ,提高衰老模型小鼠的IL 2含量 ,抗御自由基及炎症损伤 ,从而起到提高衰老模型小鼠的学习记忆能力 ,延缓脑老化的作用
To explore the mechanism of action of free radicals and immune inflammation in delaying brain aging by Alive and its alcohol extracts, and to provide experimental basis for delaying aging of the body. Ten-month-old Kunming mice were randomly divided into 5 groups. The D-galactose brain aging model was used for 8 weeks. The water maze test was used to determine the learning and memory abilities. Phosphorus method and thin layer chromatography were used to determine the cerebral cortex, hippocampus and striae. The biofilm phospholipid composition of the skeletal body was determined by radioimmunoassay and the serum IL 2 content was determined. The results showed that the number of mistakes in the water maze experiment was significantly increased in each group of mice (P < 0.01), and the maze test results in the Duhuo group were significantly lower than the model control group (P <0 01); in the cerebral cortex In the hippocampus and striatum, the PC content in the model group significantly decreased, the SM content increased, and the membrane aging index SM/PC increased significantly. After the administration, the PC content increased, the SM content decreased, SM/PC decreased, and the model control group decreased. The IL-2 content in mice was significantly lower than that in the normal control group (P < 0 01), and the results in the singaporetic drug group were significantly higher than those in the model control group (P <0 01).
