我们按照Swaiman氏方法建立胚鼠大脑皮层神经细胞原代分离单层培养。细胞可在体外存活20天以上并具有特异的形态特点及3H—GABA摄取功能。如将细胞接种到16毫米直径小皿中在体外存活后,可获得较大数量而条件近似的样品而适宜于神经生化和神经药理的研究。对于进一步在细胞和分子水平上研究发育中的脑的问题而言,这种类型的神经细胞培养具有一定实用价值。
According to the Swaiman’s method, we established the primary cultured monolayer culture of embryonic rat cerebral cortical neurons. Cells survive more than 20 days in vitro and have specific morphological features and 3H-GABA uptake function. If cells were seeded into 16 mm diameter dishes and survived in vitro, larger numbers of samples with similar conditions were obtained and were suitable for neurobiochemical and neuropharmacological studies. This type of neuronal cell culture has some practical value for further studying the developing brain at the cellular and molecular level.
