当年8月份至次年3月份采集紫花含笑(♀)×钙土含笑(♂)杂种F1代优良单株的顶芽和侧芽,置于H+0.25mg·L-16-BA+0.125mg·L-1NAA+0.10mg·L-1KT+0.05mg·L-1IAA上培养,不定芽诱导率可达115%;诱导的不定芽在H+0.3mg·L-16-BA+0.3mg·L-1KT培养基中增殖及生长较好。经过壮苗后的芽条在H+2.8mg·L-1NAA上的生根率可达100%,且单株根数多,芽条生长健壮。间苯三酚(phloroglucinol)明显促进根的发生,但对根的后期生长有一定的抑制效应。组培苗移栽至红土:腐殖土:珍珠岩=1:1:1的混合基质中,60d时成活率可达90%左右。
From August to March of the same year, the top buds and lateral buds of F1 hybrids were collected from H + 0.25 mg · L-16-BA + 0.125 mg · L -1 NAA + 0.10mg · L-1KT + 0.05mg · L-1IAA, the induction rate of adventitious buds was up to 115%. The induced adventitious buds were cultured on H + 0.3mg · L-16-BA + 0.3mg · L-1KT Medium proliferation and growth is better. After strong seedling buds in H +2.8 mg · L-1NAA on the rooting rate of up to 100%, and the number of roots per plant, shoot growth robust. Phloroglucinol obviously promoted the root development, but had certain inhibitory effect on the late growth of roots. Tissue culture seedlings transplanted to the laterite: humus: perlite = 1: 1: 1 mixed matrix, 60d survival rate of up to 90%.
