目的研究宫颈癌细胞整体甲基化水平与放疗敏感性的关系。方法用甲基转移酶抑制剂DAC处理宫颈癌He La细胞系,然后给予不同剂量放疗照射,用克隆形成实验评价He La细胞的放疗敏感性。结果 2.5和5μM浓度DAC处理过的He La细胞放疗敏感性高于未处理的He La细胞(P<0.01)。5μM浓度DAC处理过的He La细胞放疗敏感性高于2.5μM浓度DAC处理过的He La细胞(P<0.01)。结论 DAC对宫颈癌He La细胞系的放疗敏感性具有增强作用。
Objective To study the relationship between the overall methylation level of cervical cancer cells and radiosensitivity. Methods Cervical cancer He La cell line was treated with methyltransferase inhibitor DAC and irradiated with different doses of radiotherapy. The radiosensitivity of HeLa cells was evaluated by colony formation assay. Results The radiosensitivity of Hela cells treated with 2.5 and 5 μM DAC was higher than that of untreated He La cells (P <0.01). Hela cells treated with 5μM DAC had higher radiosensitivity than Hela cells treated with 2.5μM DAC (P <0.01). Conclusion DAC enhances the radiosensitivity of cervical cancer He La cell line.
