设置6个不同光照和温度的处理组合,研究温度和光照条件对粉蕉组培快繁的影响,探索一套适合‘金粉1号’粉蕉的组培快繁新技术。结果表明,在继代培养阶段,高温暗培养处理继代芽萌动早、分化早、分化快,继代培养25 d时,繁殖系数2.52,有效芽率96.74%,高于其他处理,发泡芽率5.43%,显著低于其他处理;生根培养阶段,除常温光培养处理外,高温暗培养处理等处理平均每株苗根数均大于5条,假值成活率高。综合考虑繁殖系数、分化效率、继代苗和生根苗质量、节能等多方面因素,粉蕉组培快繁时,高温暗培养处理组合最优。
Six treatment combinations with different light and temperature were set up to study the effect of temperature and light on the tissue culture and propagation of S. japonica and to explore a set of new rapid propagation techniques for tissue culture of ’Jinfen No.1’. The results showed that in the subculture stage, the germination rate of the secondary germination was earlier, the differentiation was earlier and the differentiation was faster, the propagation coefficient was 2.52 and the effective germination rate was 96.74% when subcultured for 25 days, which was higher than other treatments. The rate of 5.43% was significantly lower than that of other treatments. In the rooting stage, the average number of shoots per plant was more than 5 and the false survival rate was high except for the light culture at room temperature. Considering propagation coefficient, differentiation efficiency, succession seedling and rooting seedling quality, energy saving and other factors, the best combination of high temperature and dark culture was obtained when the plantlets were rapidly cultivated.
