目的:观察中药复方联合三苯氧胺(TAM)对MCF-7乳腺癌的抑制作用及形态学的影响。方法:将造模成功小鼠随机分为对照组、TAM组、乳结合组(乳岩宁+TAM)和益结合组(益气消积方+TAM)。日一次灌胃,连续21 d,实验前后测量裸鼠体重变化情况;用药21天后剥离肿瘤称重,计算抑瘤率;HE染色后光镜下观察瘤组织病理形态学变化;并透射电镜(TEM)下观察超微结构变化。结果:体重:TAM组裸鼠体重下降最明显(3.18±0.061),结合组次之(2.5±0.042),二者比较显著意义(P<0.01);瘤重:TAM组、乳结合组、益结合组的的肿瘤重量均低于对照组,与对照组比较有统计学差异(P<0.01);HE染色提示乳腺癌为浸润性导管癌,透射电镜下观察提示中药复方联合TAM具有诱导肿瘤细胞凋亡的作用。结论:中药复方联合TAM能抑制肿瘤生长,增加TAM抑瘤率,其作用机制可能通过诱导肿瘤细胞凋亡来实现的。
Objective: To observe the inhibitory effect of traditional Chinese medicine combined with tamoxifen (TAM) on MCF-7 breast cancer and its morphological changes. Methods: The successful mice were randomly divided into control group, TAM group, milk-binding group (YURAN + TAM), and Yixi-combination group (YQI + TAM). The tumor weight was measured before and after the experiment. The tumors were weighed after 21 days of treatment and the tumor inhibition rate was calculated. The histopathological changes of the tumor tissues were observed under light microscope after HE staining. ) Observed ultrastructural changes. Body weight: The body weight of the TAM group was significantly lower than that of the TAM group (3.18 ± 0.061) and that of the combined group (2.5 ± 0.042) (P <0.01) The tumor weight of the combination group was lower than that of the control group, and there was a significant difference compared with the control group (P <0.01). HE staining showed that the invasive ductal carcinoma of breast cancer was observed by transmission electron microscopy. The role of apoptosis. Conclusion: TAM combined with traditional Chinese medicine can inhibit tumor growth and increase tumor inhibitory rate of TAM. The mechanism may be through inducing tumor cell apoptosis.
