目的探讨虎杖苷对大鼠脑出血性损伤的治疗作用及作用机制。方法采用Ⅶ型胶原酶诱导大鼠脑出血,建立脑出血模型,观察虎杖苷对大鼠神经功能症状、脑组织中超氧化物歧化酶(SOD)活力、丙二醛(MDA)水平,脑组织Bcl-2蛋白表达,及血清中白细胞介素-1β(IL-1β)水平的影响。结果与脑出血模型组相比,虎杖苷(25、50、100 mg/kg)可显著改善脑出血后神经功能缺损,提高脑组织SOD活力、降低MDA水平,增加Bcl-2蛋白表达,降低血清中IL-1β水平,其作用与剂量呈正相关。结论虎杖苷通过对抗自由基损伤,抗凋亡及抗炎等作用,发挥对大鼠脑出血损伤的治疗作用。
Objective To investigate the therapeutic effect and mechanism of polydatin on cerebral hemorrhagic injury in rats. Methods Cerebral hemorrhage induced by type VII collagenase was used to establish a cerebral hemorrhage model. The neurological symptoms, superoxide dismutase (SOD) activity and malondialdehyde (MDA) levels in rat brain were observed with polydatin. The brain tissue was Bcl. -2 protein expression, and the influence of serum interleukin-1β (IL-1β) levels. Results Compared with the cerebral hemorrhage model group, polydatin (25, 50, 100 mg/kg) significantly improved neurological deficits after cerebral hemorrhage, increased brain SOD activity, decreased MDA levels, increased Bcl-2 protein expression, and decreased serum levels. The level of IL-1β was positively correlated with the dose. Conclusion Polydatin played an important role in the treatment of cerebral hemorrhage in rats by fighting free radical damage, anti-apoptotic and anti-inflammatory effects.
