对两脱胶菌株Ke34及Le10的生长繁殖条件进行了测试。结果表明,它们的适宜扩繁培养基以豆饼粉培养基为最佳、其次为麦芙皮培养基,其中豆饼粉的含量以3%为宜;最适培养温度为35~40℃之间;最适pH值为70(中性);但是,以3%的豆饼粉培养基,pH值取其自然,培养效果也很理想。在优选各项测试的条件下,测定它们的生长曲线,并结合涂片镜检观察菌体变化,最终确定两脱胶菌株Ke34及Le10,以3%的豆饼粉为扩繁培养基、pH值70或取其自然,在有氧的条件,培养16~24h即为接种沤麻脱胶的最佳时间。
The growth and reproduction conditions of the two degummed strains Ke34 and Le10 were tested. The results showed that the suitable culture medium for soybean flour was the best, followed by the wheat flour medium, in which the content of bean flour was 3%, the optimum culture temperature was 35-40 ℃, The optimum pH value is 7.0 (neutral); however, with 3% soybean meal medium, the pH value is natural and the culture effect is also very good. Under the optimal conditions, the growth curves were measured and microscopic observation of bacterial cells with smear microscopy showed that the two de-gluten strains Ke34 and Le10 were finally identified.
