目的观察重组人白细胞介素(rhIL)-15对放射线照射的T淋巴细胞损伤的保护作用。方法从30例正常人的骨髓分离淋巴细胞,随机分为实验组及对照组各15例。实验组加入100μg/L的IL15,两组分别放射线照射,流式细胞术和细胞核形态检测等方法观察照射前后T细胞亚群、bclXL、bax和bcl-2蛋白水平变化和淋巴细胞凋亡率。结果放射线照射后淋巴细胞出现大量凋亡;实验组的淋巴细胞凋亡率较对照组明显降低,T细胞亚群表达率较对照组明显升高,差异有统计学意义(P<0.01);实验组的细胞凋亡相关基因bcl2、bclXL的表达水平较对照组上调,bax表达则较对照组下调(P<0.01)。结论IL-15能抑制放射线引起的淋巴细胞凋亡,改善急性放射病引起的免疫损伤。
Objective To observe the protective effect of recombinant human interleukin (IL-15) on radiation-induced T lymphocyte injury. Methods Lymphocytes were isolated from bone marrow of 30 normal subjects and randomly divided into experimental group and control group, 15 cases each. The experimental group was given 100μg / L IL15, and the changes of T cell subsets, the expression of bclXL, bax and bcl-2 and the rate of lymphocyte apoptosis were observed before and after irradiation by radiation, flow cytometry and nuclear morphology. Results A large number of apoptotic lymphocytes appeared after irradiation. The apoptosis rate of lymphocytes in experimental group was significantly lower than that in control group, and the expression of T lymphocyte subsets was significantly higher than that in control group (P <0.01) The expressions of bcl2 and bclXL were upregulated compared with the control group, while the expression of bax was down-regulated compared with the control group (P <0.01). Conclusion IL-15 can inhibit radiation-induced apoptosis of lymphocytes and improve immune damage induced by acute radiation sickness.
