目的:观察泄浊解毒方对溃疡性结肠炎(UC)大鼠肠黏膜上皮细胞(IECs)TLR2基因、蛋白表达的影响。方法:40只健康清洁级SD大鼠随机分5组:空白组,模型组,柳氮磺胺吡啶组,泄浊解毒方大、小剂量组,除正常组外采用2,4,6-三硝基苯磺酸(TNBS)/乙醇混合溶液灌肠复制UC大鼠模型,造模成功后,各治疗组大鼠灌服相应药物,正常组及模型组予0.9%氯化钠溶液灌胃。2周后处死全部大鼠,RT-PCR法检测TLR2基因表达,免疫组化法检测TLR2蛋白表达。结果:泄浊解毒方各剂量组大鼠TLR2基因、蛋白表达均明显低于模型组;泄浊解毒方大剂量组TLR2 mRNA、蛋白表达明显低于柳氮磺胺吡啶组及小剂量组。结论:泄浊解毒方对大鼠UC有良好的治疗作用,其作用机制可能与降低TLR2基因、蛋白表达有关。
Objective: To observe the effect of Xiezhuojiedu Formula on TLR2 gene and protein expression in intestinal mucosal epithelial cells (IECs) of rats with ulcerative colitis (UC). Methods: Forty healthy SD rats were randomly divided into 5 groups: blank group, model group, sulfasalazine group, Xiezhuojiedu Fangda group and low dose group. Except normal group, 2,4,6-trinitrobenzene After the model was successfully established, the rats in each treatment group were drenched with the corresponding drugs, and the normal group and the model group were orally administered with 0.9% sodium chloride solution. After 2 weeks, all rats were killed, TLR2 gene expression was detected by RT-PCR and TLR2 protein by immunohistochemistry. Results: The expressions of TLR2 mRNA and protein were significantly lower in the Xiezuojiedu Fang group than those in the untreated group. The expression of TLR2 mRNA and protein in the Xiezuojiedu Fang high dose group was significantly lower than that in the sulfasalazine group and the low dose group. Conclusion: Xiezhuojiedu decoction has a good therapeutic effect on UC in rats, and its mechanism may be related to the decrease of TLR2 gene and protein expression.
