目的探讨猪苓汤及泽泻对肾结石大鼠骨桥蛋白(OPN)mRNA表达的影响。方法采用乙醛酸溶液制作大鼠肾结石模型,用反转录聚合酶链反应(RT-PCR)技术检测肾结石大鼠OsteopotinmRNA的表达。结果诱石剂可使大鼠肾脏OPNmRNA的表达量明显增加(P<0.05);而猪苓汤及泽泻则可抑制OPNmRNA的表达(P<0.05)。结论初步揭示猪苓汤及泽泻对肾结石大鼠OsteopotinmRNA表达的变化规律,从而为临床疾病的防治及治疗提供一定的依据。
Objective To investigate the effects of Zhuling Decoction and Alisma on the expression of osteopontin (OPN) mRNA in rats with kidney stone. Methods Rat kidney stones models were made with glyoxylic acid solution. The expression of Osteopotin mRNA in kidney stones rats was detected by reverse transcription-polymerase chain reaction (RT-PCR). Results The amount of OPN mRNA in the rat kidney was significantly increased by the stone attractant (P<0.05), while the pig’s soup and Alisma could inhibit the expression of OPN mRNA (P<0.05). Conclusion The changes of Osteopotin mRNA expression in rats with kidney stones were initially revealed, which provided a basis for the prevention and treatment of clinical diseases.
