目的 :观察华蟾素对细胞生长及凋亡的影响。方法 :应用体外细胞培养技术、MTT测定、集落形成试验、电镜观察及DNA琼脂糖凝胶电泳等方法。结果 :华蟾素对MGC 80 3细胞的生长有显著抑制作用 ,能抑制细胞的集落形成 ,MTT法测定 ,其IC50 约为 0 16 μg/ml;电镜下观察到细胞呈凋亡特征 ;琼脂糖凝胶电泳显示DNA呈梯状降解。 结论 :华蟾素能抑制MGC- 80 - 3细胞生长 ,药物作用方式在较高浓度时以细胞毒作用为主 ,在较低浓度时以诱导细胞凋亡为主。诱导细胞凋亡是华蟾素抗肿瘤的重要方式之一。
Objective: To observe the effects of cinobufacini on cell growth and apoptosis. Methods: In vitro cell culture technique, MTT assay, colony formation assay, electron microscopy and DNA agarose gel electrophoresis were used. Results: Cinobufacini significantly inhibited the growth of MGC 80 3 cells and inhibited the colony formation. The IC50 of the cells was about 0 16 μg / ml. The apoptosis of cells was observed under the electron microscope. The agarose Gel electrophoresis showed that the DNA was degraded in a ladder manner. CONCLUSION: Cinobufacini inhibits the growth of MGC-80 - 3 cells. The cytotoxicity of the drug at higher concentrations is mainly induced by cinobufacini, and the apoptosis is mainly induced at lower concentration. Induction of apoptosis is one of the most important anti-tumor strategies for cinobufacini.
