目的探讨小鼠免疫器官胸腺、脾脏及淋巴结组织中Fgl2凝血酶原酶基因转录及蛋白表达水平。方法使用RT-PCR分析Fgl2凝血酶原酶基因转录水平;免疫组化检测淋巴器官标本Fgl2凝血酶原酶表达;双色免疫荧光分析Fgl2凝血酶原酶与Foxp3表达的关系。结果RT-PCR结果证实正常的胸腺、脾脏及淋巴结有Fgl2凝血酶原酶基因转录。免疫组化的结果则表明,这些淋巴器官中有大量的Fgl2凝血酶原酶表达。免疫荧光分析结果显示胸腺及脾脏内的Fgl2~+细胞为Foxp3~-细胞。结论免疫器官内Fgl2凝血酶原酶的表达可能参与了T细胞发育及功能。此外,Fgl2不直接参与Foxp3~+调节性T细胞的抑制功能。
Objective To investigate the gene transcription and protein expression of Fgl2 prothrombinase in the thymus, spleen and lymph node of the immune organs of mice. Methods The transcription level of Fgl2 prothrombinase gene was analyzed by RT-PCR. The expression of Fgl2 prothrombinase was detected by immunohistochemistry. The relationship between Fgl2 prothrombinase and Foxp3 expression was analyzed by two-color immunofluorescence. Results RT-PCR confirmed that normal thymus, spleen and lymph nodes Fgl2 prothrombinase gene transcription. Immunohistochemical results show that these lymphoid organs have a large number of Fgl2 prothrombinase expression. Immunofluorescence analysis showed that Fgl2 ~ + cells in the thymus and spleen were Foxp3 ~ - cells. Conclusion The expression of Fgl2 prothrombinase in immune organs may be involved in the development and function of T cells. In addition, Fgl2 is not directly involved in Foxp3 ~ + regulatory T cell suppressive function.
