目的:研究CD24基因单核苷酸多态性(SNP)位点rs52812045与中国汉族人群系统性红斑狼疮(SLE)的相关性。方法:采用聚合酶链反应及限制性片段长度多态性(PCR-RFLP)方法,对54例SLE患者及70例健康对照者CD24SNP位点rs52812045进行多态性检测。结果:CD24基因位点rs52812045可以检测到T/T、T/C、C/C 3种等位基因分布;系统性红斑狼疮患者和健康对照者CD24基因型T/T的频率分别为29.6%和11.5%,CD24的杂合基因型T/C的分布分别为51.9%和31.4%,基因型C/C的频率分别为18.5%和57.1%。统计学分析,SLE组CD24基因rs52812045位点T等位基因携带者显著高于正常对照组[分别为73.1%和42.9%,χ2=9.318,P<0.05,OR=2.182(1.068-4.458)],T等位基因频率显著高于对照组(分别为44.4%和27.1%,χ2=8.056,P<0.05)。结论:CD24SNP位点rs52812045与中国汉族人群SLE的发病具有显著相关性;CD24SNP位点rs52812045的T等位基因可能是SLE的易感基因之一。
Objective: To investigate the association between SNP rs52812045 of CD24 gene and systemic lupus erythematosus (SLE) in Chinese Han population. Methods: Polymorphism of rs52812045 in CD24SNP locus was detected in 54 SLE patients and 70 healthy controls by polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP). Results: The T / T, T / C and C / C allele distributions were detected at rs 5258120 of CD24 locus. The T / T frequencies of CD24 genotypes in patients with systemic lupus erythematosus and healthy controls were 29.6% and 11.5%, CD24 heterozygous genotype T / C distribution were 51.9% and 31.4%, genotype C / C frequency were 18.5% and 57.1%. Statistical analysis, SLE group CD24 gene rs52812045 T allele carriers were significantly higher than the control group [73.1% and 42.9%, respectively, χ2 = 9.318, P <0.05, OR = 2.182 (1.068-4.458] T allele frequency was significantly higher than the control group (44.4% and 27.1%, respectively, χ2 = 8.056, P <0.05). CONCLUSIONS: There is a significant correlation between the CD24 SNP rs52812045 and the incidence of SLE in Chinese Han population. The T allele of rs245812045 at CD24 SNP may be one of the susceptible genes of SLE.
