目的:研究姜黄素对增生性瘢痕成纤维细胞增殖的作用,以期为姜黄素用于增生性瘢痕的治疗提供实验依据。方法:以增生性瘢痕成纤维细胞(HSFBs)和正常皮肤成纤维细胞(NSFBs)作为工具细胞,分别通过MTT细胞毒性试验、细胞生长曲线和蛋白印记法分析姜黄素对细胞的增殖影响。结果:姜黄素能够抑制HSFBs细胞的增殖而姜黄素对NSFBs的抑制作用相对较小;在姜黄素处理后NSFBs细胞p-JNK蛋白表达上调,使用JNK抑制剂后,能阻断JNK蛋白的磷酸化,并且降低姜黄素诱导细胞凋亡的能力。结论:姜黄素能够选择性的抑制HSFBs细胞的增殖,或可成为治疗增生性瘢痕的药物。
Objective: To study the effect of curcumin on the proliferation of hypertrophic scar fibroblasts in order to provide experimental evidence for the treatment of hypertrophic scars with curcumin. Methods: Hyperplastic scar fibroblasts (HSFBs) and normal skin fibroblasts (NSFBs) were used as tool cells. The effects of curcumin on cell proliferation were analyzed by MTT cytotoxicity assay, cell growth curve and Western blot respectively. Results: Curcumin inhibited the proliferation of HSFBs cells and the inhibitory effect of curcumin on NSFBs was relatively small. After curcumin treatment, the expression of p-JNK protein was up-regulated in NSFBs cells. JNK inhibitor could block the phosphorylation of JNK protein , And reduces the ability of curcumin to induce apoptosis. CONCLUSION: Curcumin can selectively inhibit the proliferation of HSFBs cells or become a drug for the treatment of hypertrophic scars.
