建立测定赤芝中灵芝酸A的HPLC方法,并研究赤芝不同生长期和不同部位灵芝酸A的含量。Agilent Zorbax SB-C18色谱柱(250 mm×4.6 mm,5 m),乙腈(A)-0.05 mol·L~(-1)磷酸二氢铵水溶液(B)作为流动相梯度洗脱,体积流量1.0 mL·min~(-1),柱温25℃,检测波长254 nm。结果表明,灵芝酸A在0.6165 mg~8.16 mg范围内有良好的线性关系;平均回收率为96.1%(n=9)。不同生长期和不同部位灵芝酸A含量有差异:未成熟的灵芝蕾>成熟期的子实体,菌盖>柄。通过对不同生长期和不同部位灵芝酸A的含量比较分析,为建立赤芝质量标准和合理开发利用提供了参考依据。
To establish a HPLC method for the determination of Ganoderma lucidum A in Ganoderma lucidum, and to study the content of Ganoderma lucidum A in different growth stages and different parts of Ganoderma lucidum. Agilent Zorbax SB-C18 column (250 mm × 4.6 mm, 5 m), acetonitrile-A (0.05 mol·L -1) ammonium dihydrogen phosphate aqueous solution (B) as mobile phase gradient elution, mL · min -1, the column temperature was 25 ℃ and the detection wavelength was 254 nm. The results showed that Ganoderma acid A had a good linear relationship in the range of 0.6165 mg ~ 8.16 mg with an average recovery of 96.1% (n = 9). Different growth stages and different parts of Ganoderma lucidum acid A content differences: immature Ganoderma Lei> mature fruiting body, cap> handle. By comparing the contents of Ganoderma lucidum A in different growth stages and different parts, it provides a reference for establishing the quality standard and developing and utilizing the Ganoderma lucidum.
