目的:探讨通络保肾复方对肾小球脏层上皮细胞(POD)增殖反应及转分化生长因子-β1(TGF-β1)mRNA表达的影响。方法:将5%含药血清培养基加入体外培养的足细胞,分别于高糖环境下脂多糖(LPS)刺激足细胞12h、24h、36h、48h后,用MTT方法检测各组的增殖情况,RT-PCR方法检测TGF-β1mRNA的表达。结果:LPS刺激12h后足细胞开始增殖,24h达高峰,48h后开始下降。通络保肾复方能够抑制足细胞增殖,以通络保肾复方大剂量组效果显著,与LPS组比较差异显著(P<0.01)。同时通络保肾复方可以降低足细胞TGF-β1mRNA的表达。本研究结果提示,通络保肾复方可以抑制高糖环境下LPS刺激诱导的足细胞增殖及降低足细胞TGF-β1mRNA的表达,减轻足细胞的损伤,抑制细胞外基质的异常积聚和沉积,从而延缓肾小球硬化的进程。
Objective: To investigate the effect of Tongluo Baoshen compound on the proliferation of glomerular visceral epithelial cells (POD) and the expression of transdifferentiated growth factor-β1 (TGF-β1) mRNA. METHODS: 5% drug-containing serum medium was added to podocytes cultured in vitro, and podocytes were stimulated with lipopolysaccharide (LPS) for 12 h, 24 h, 36 h, and 48 h in a high glucose environment. The proliferation of each group was detected by MTT assay. The expression of TGF-β1 mRNA was detected by RT-PCR. RESULTS: After 12h of LPS stimulation, podocytes began to proliferate, peaked at 24h and began to decline after 48h. Tongluo Baoshen compound can inhibit the proliferation of podocytes, and the effect of Tongluo Baoshen compound high-dose group is significant, which is significantly different from that of LPS group (P<0.01). At the same time, Tongluo Baoshen compound can reduce the expression of TGF-β1 mRNA in podocytes. The results of this study suggest that Tongluo Baoshen compound can inhibit the proliferation of podocytes induced by LPS stimulation in high glucose environment and reduce the expression of TGF-β1 mRNA, reduce podocyte injury and inhibit abnormal accumulation and deposition of extracellular matrix. Delay the progression of glomerulosclerosis.
