目的 研究三氧化二砷对猪髂动脉内皮细胞 (PIEC)的诱导凋亡作用。方法 采用MTT、细胞生长曲线、倒置相差显微镜、细胞爬片HE染色、细胞涂片苏丹Ⅲ染色、透射电镜等观察三氧化二砷作用后PIEC的变化。结果 PIEC细胞与三氧化二砷共孵育后出现细胞变圆、贴壁减少、上浮、细胞空泡样变性 ,脂肪颗粒增多 ;电镜等观察到核膜皱缩、核染色质凝结、边集等细胞凋亡现象 ,并且在一定的范围内该现象随药物浓度的增加而明显。结论 一定浓度的三氧化二砷能抑制PIEC细胞的生长 ,细胞脂肪变性 ,并诱导细胞凋亡。提示向肿瘤组织局部导入三氧化二砷可以抑制肿瘤局部供血动脉内皮的生长 ,促进供血动脉闭塞。
Objective To study the apoptotic effect of arsenic trioxide on Porcine iliac artery endothelial cells (PIEC). Methods The changes of PIEC were observed with MTT, cell growth curve, inverted phase contrast microscopy, cell slide HE staining, cell smear Sudan III staining, and transmission electron microscopy. Results After co-incubation of PIEC cells with arsenic trioxide, cell rounding, adherence decrease, floating, cell vacuolar degeneration and fat particles increased; Electron microscopy and other observations of nuclear membrane shrinkage, nuclear chromatin condensation, edge collection and other cell apoptosis phenomenon , And in a certain range, this phenomenon is obvious with the increase of drug concentration. CONCLUSION: A certain concentration of arsenic trioxide can inhibit the growth of PIEC cells, induce cell steatosis, and induce apoptosis. It is suggested that local introduction of arsenic trioxide into the tumor tissue can inhibit the growth of the local blood supply arterial endothelium and promote the occlusion of the supplying arteries.
