目的探讨不同剂量雷公藤多苷(TWP)对糖尿病大鼠肾脏的保护作用。方法高糖高脂喂养联合小剂量STZ(30 mg/kg)构建T2DM大鼠模型,予不同剂量TWP治疗8周,测定大鼠24 hUAlb及血液生化指标,并观察肾脏病理变化,检测肾脏nephrin、podocin和血管内皮生长因子(VEGF)表达水平的变化。结果 DM组24 hUAlb高于NC组[(253.7±53.0)vs(45.7±14.5)μg/24 h];与DM组比较,TWP干预后,24 hUAlb减少[(253.7±53.0)vs(183.5±78.7)、(123.8±74.8)、(119.0±52.0)μg/24 h],TWP可改善糖尿病大鼠肾脏病理改变,并使nephrin和podocin表达量增高,VEGF表达量下降。结论 TWP通过上调nephrin和podocin及下调VEGF的表达,对糖尿病大鼠肾脏起到保护作用。
Objective To investigate the protective effects of different doses of tripterygium wilfordii glycosides (TWP) on the kidney of diabetic rats. Methods High glucose and high fat diet combined with low dose of STZ (30 mg / kg) was used to establish T2DM model. The rats were treated with different doses of TWP for 8 weeks. The serum levels of 24 hUAlb and blood biochemical parameters were measured. The renal pathological changes, nephrin, podocin and vascular endothelial growth factor (VEGF) expression levels. Results Compared with DM group, 24 hUAlb in DM group was significantly lower than that in NC group [(253.7 ± 53.0) vs (45.7 ± 14.5) μg / 24 h] ), (123.8 ± 74.8) and (119.0 ± 52.0) μg / 24 h, respectively. TWP could improve renal pathological changes in diabetic rats and increase nephrin and podocin expression and decrease VEGF expression. Conclusion TWP can protect the kidney of diabetic rats by up-regulating nephrin and podocin and down-regulating the expression of VEGF.
