目的研究17β雌二醇(17β-estradiol,E2)对核转录因子(NF-кB)转录活性及Toll样受体(TLR4)表达的影响。方法采用脂多糖和17β雌二醇同时作用于单核巨噬细胞RAW264.7,采用逆转录-聚合酶链反应方法检测细胞表面TLR4基因的表达;用荧光素酶报告基因转染试验观察NF-кB转录活性变化。结果对照组、LPS单独刺激组和17β雌二醇预作用组细胞TLR4 mRNA的相对表达分别为(1.23±0.21)(、1.52±0.28)和(1.11±0.27),NF-κB转录活性的相对值分别为(126±32)(、306±58)和(247±37),LPS刺激可使细胞TLR4 mRNA表达和NF-кB转录活性增强,而17β雌二醇预作用可明显降低TLR4 mRNA的表达和NF-кB转录活性,组间比较差异具有统计学意义(P<0.05)。结论 17β雌二醇能通过调节TLR4及转录因子NF-кB的活性呈现一定的抗炎作用。
Objective To investigate the effect of 17β-estradiol (E2) on the transcriptional activity of nuclear factor-kappa B (NF-κB) and the expression of Toll-like receptor (TLR4). Methods Lipopolysaccharide and 17β-estradiol were applied simultaneously to RAW264.7 monocyte-macrophage cells. The TLR4 gene expression was detected by reverse transcription-polymerase chain reaction (RT-PCR) кB transcriptional activity changes. Results The relative expressions of TLR4 mRNA in control group, LPS alone group and 17β estradiol preconditioning group were (1.23 ± 0.21) (, 1.52 ± 0.28) and (1.11 ± 0.27), respectively, and the relative values of NF-κB transcriptional activity (126 ± 32), (306 ± 58) and (247 ± 37), respectively. LPS stimulated the expression of TLR4 mRNA and NF-κB transcriptional activity, while 17β estradiol pretreatment significantly decreased the expression of TLR4 mRNA And NF-кB transcription activity, the difference between the groups was statistically significant (P <0.05). Conclusion 17β-estradiol can exert a certain anti-inflammatory effect by regulating the activity of TLR4 and the transcription factor NF-кB.
