目的 探讨恒磁场对离体兔主动脉平滑肌细胞增殖的抑制作用 ,为研究冠脉内磁化支架对经皮腔内冠脉成形术 (PTCA)及支架术后冠脉再狭窄的防治作用提供理论基础。方法 用含10 %小牛血清的RPMI - 16 40培养液体外培养新西兰白兔的主动脉平滑肌细胞至 3~ 7代 ,将含 5 %小牛血清的RPMI- 16 40培养液主动脉平滑肌细胞悬液 10 0 μl加入 96孔培养板中培养 ,对照组持续培养 72h ,实验组 2 4h后将培养板置于 0 .35T的恒磁场中继续培养 48h ,两组培养 72h后用MTT法和3 H -TdR掺入法检测动脉平滑肌细胞的增殖数量 (A值和核素闪烁计数值 )。结果 在恒磁场的作用下 ,实验组主动脉平滑肌细胞的增殖数量 (0 .2 948± 0 .0 44 8和 1378± 5 96 )min ,显著低于对照组平滑肌细胞的增殖数量 (0 .4783± 0 .0 5 80和 2 6 87± 983)min ,P <0 .0 0 1。结论 恒磁场对动脉平滑肌细胞的增殖有显著的抑制作用 ;磁场对PTCA支架后冠脉再狭窄可能具有防治作用
Objective To investigate the inhibitory effect of constant magnetic field on the proliferation of isolated rabbit aortic smooth muscle cells in order to provide theoretical basis for the study on the prevention and treatment of intracoronary magnetized stents on percutaneous transluminal coronary angioplasty (PTCA) and coronary stenting . Methods Aorta smooth muscle cells of New Zealand white rabbits were cultured in vitro with RPMI - 1640 medium containing 10% fetal bovine serum for three to seven generations. The aortic smooth muscle cells suspended in 5% fetal bovine serum (RPMI-1640) Liquid 10 0 μl was added to 96-well culture plate, the control group continued to culture 72h, experimental group 2 4h after the plate was placed in 0.35T constant magnetic field for 48h, two groups were cultured for 72h MTT method and 3 H -TdR incorporation method to detect the proliferation of arterial smooth muscle cells (A value and scintillation count). Results The number of proliferation of aortic smooth muscle cells (0.2948 ± 0.0448 and 1378 ± 5 96) min in the experimental group was significantly lower than that in the control group under the action of constant magnetic field (0.4783 ± 0. 0 5 80 and 2 6 87 ± 983) min, P <0 0 01. CONCLUSION: The constant magnetic field can significantly inhibit the proliferation of arterial smooth muscle cells. Magnetic field may play a preventive and therapeutic role in the restenosis of coronary artery after PTCA
