本研究旨在提高基因转染人红白血病细胞株K562的转染效率,同时观察绿色荧光蛋白(GFP)作为报告系统用于转染条件优化的可行性。目的基因以GFP为报告系统,通过脂质体转染及电穿孔两种方式转染K562细胞,荧光显微镜下观察并计算转染效率。结果表明:电转染较脂质体转染K562细胞的效率高,电穿孔转染效率在10%左右,而脂质体转染效率小于1%。结论:电转染较脂质体转染K562细胞的效率高,GFP可作为报告系统优化K562细胞转染条件。
The purpose of this study was to improve the transfection efficiency of gene transfection in human erythroleukemia cell line K562 and to observe the feasibility of green fluorescent protein (GFP) as a reporter system for the optimization of transfection conditions. The target gene was transfected into K562 cells by liposome transfection and electroporation using GFP as reporter system. The transfection efficiency was observed under a fluorescence microscope. The results showed that the efficiency of transfection with lipofectamine 2000 was higher than that with lipofectamine. The transfection efficiency of electroporation was about 10%, while the transfection efficiency of liposome was less than 1%. Conclusion: The efficiency of transfecting K562 cells with lipofectamine is higher than that of lipofectamine. GFP can be used as a reporter to optimize the transfection conditions of K562 cells.
