Objectives To explore the protective effect of irbesartan(Irb) under the interference with angiotensinⅡ(AngⅡ) on ABCA1.Methods Electron microscopy was used to detect the morphous of foam cells.The expression of ABCA1 mRNA and its protein were determined by RT-PCR and Western blotting,respectively.The variance of cellular cholesterol content was measured by zymochemistry via-fluorospectrophotometer.Results A positive facilitative effect of AngⅡon the formation of foam cells was observed.Total cholesterol was increased significantly by AngⅡ,the expression of ABCA1 was down-regulated obviously by AngⅡ;Irb could protect ABCA1 against the lesion of AngⅡ;Total cellular cholesterol content was reduced significantly in Irb + AngⅡgroup;However,considerable alteration about the cholesterol content and the expression of ABCA1 were not observed in Irb group without incubation with AngⅡ.Conclusions Irb could protect ABCA1 against the lesion of AngⅡ,which may contribute to its anti-atherosclerotic properties.
