目的 探讨乳腺癌组织 p2 1WAF1DNA多态性及其与预后的关系。方法 采用聚合酶链反应单链构象多态性技术 (PCR SSCP)和免疫组化S P法检测 10 0例乳腺癌组织 p2 1WAF1DNA多态性和蛋白表达 ,并根据随访结果行预后分析。结果 10 0 % (18/ 18例 )的具有 p2 1WAF1DNA多态性的乳腺癌组织 p2 1WAF1蛋白表达阳性 ,39% (32 / 82 )的无 p2 1WAF1DNA多态性乳腺癌组织蛋白表达阳性 ,p2 1WAF1DNA多态性组明显高于无多态性组 (χ2 =2 1 95 ,P <0 0 1) ;p2 1WAF1DNA多态性与p2 1WAF1蛋白表达呈正相关 (rs=0 5 76 ,P <0 0 1)。单因素预后分析显示 p2 1WAF1DNA多态性组生存期低于无多态性组 (χ2 =6 0 2 ,P <0 0 1)。结论 乳腺癌p2 1WAF1DNA多态性可能是造成乳腺癌患者生存期缩短的一个重要因素。
Objective To investigate the p2 1WAF1 DNA polymorphism in breast cancer and its relationship with prognosis. Methods PCR-SSCP and immunohistochemical SP methods were used to detect the polymorphism of p2 1WAF1 DNA in 100 cases of breast cancer. The prognosis was analyzed according to the follow-up results. Results The positive expression of p2 1WAF1 protein in breast cancer tissues with p2 1WAF1 DNA polymorphism was found in 10 0% (18 of 18) cases, and positive expression of p2 1 WAF1 DNA polymorphic breast cancer tissues in 39% (32 of 82) cases was positive The polymorphism of p2 1WAF1 was positively correlated with the expression of p2 1WAF1 (rs = 0 5 76, P 0 01 ). Univariate analysis showed that the survival of p2 1WAF1 DNA polymorphism group was lower than that of non-polymorphism group (χ2 = 6002, P <0.01). Conclusion The p2 1WAF1 DNA polymorphism in breast cancer may be an important factor in the shortened survival of patients with breast cancer.
