为了解自发性高血压大鼠 (SHR)肾损害的程度 ,通过检测SHR血浆、肾组织中内皮素 (ET)的水平、碱性成纤维细胞生长因子 (bFGF)的表达 ,初步探讨SHR肾损害的机制。将 1 6个月龄SHR作为实验组 ,同龄WKY大鼠作为对照组 ;检测CCr、UAE ;采用放射免疫分析法测定大鼠血浆、肾组织中ET的水平 ;肾组织切片采用HE染色 ,PAS染色 ,用免疫组化检测Ⅰ、Ⅳ型胶原和bFGF蛋白的表达 ,用原位杂交方法检测肾组织中bFGFmRNA的表达。结果显示 ,SHR肾功能损害明显 ,肾组织病理改变明显 ,存在肾小球硬化及间质纤维化 ;SHR血浆、肾组织ET含量明显升高 ;肾组织中bFGF的表达明显增强。提示ET及bFGF可能参与了SHR肾损害
To investigate the degree of renal damage in spontaneously hypertensive rats (SHR), SHR renal damage was investigated by detecting the level of endothelin (ET) and the expression of basic fibroblast growth factor (bFGF) in SHR plasma and kidney tissue Mechanisms. The SHR of 16 months old was used as the experimental group and the WKY rats of the same age as the control group. The CCr and UAE were detected. The levels of ET in the plasma and kidney tissue of rats were determined by radioimmunoassay. The renal tissue sections were stained with HE and PAS The expressions of type Ⅰ collagen and bFGF protein were detected by immunohistochemistry. The expression of bFGF mRNA in renal tissues was detected by in situ hybridization. The results showed that the renal damage of SHR was obvious, the pathological changes of renal tissue were obvious, there was glomerulosclerosis and interstitial fibrosis; the content of ET in plasma and kidney of SHR was obviously increased; the expression of bFGF in renal tissue was obviously increased. It is suggested that ET and bFGF may participate in SHR renal damage
