用壳聚糖与CuSO2.5H2O溶液络合成的壳聚糖铜为载体,固定化白毒鹅膏菌胞外漆酶,测定了铜盐浓度、络合时间、固定化时缓冲液的pH值、固定化时间、给酶量对固定化酶性质的影响。实验得出固定化最佳条件是在20℃、pH7.0的磷酸缓冲液中,固定2个小时,这种条件下酶活保持率为50.2%。固定化酶的最适pH值4.0比游离酶的4.6偏酸;固定化酶的最适温度24℃。
The chitosan copper complexed with CuSO2.5H2O solution was used as the carrier to immobilize extracellular laccase of Amanita auricula. The effects of copper salt concentration, complexation time, pH of the buffer solution , Immobilization time, the amount of enzyme on the nature of the immobilized enzyme. The experimental results showed that the optimum conditions for immobilization were immobilized in phosphate buffer solution of pH7.0 at 20 ℃ for 2 hours under the conditions of 50.2% enzyme activity. The optimum pH value of immobilized enzyme 4.0 is 4.6 more than that of free enzyme. The optimum temperature of immobilized enzyme is 24 ℃.
