目的:通过观察石斛合剂(DC)对链脲佐菌素(STZ)损伤的胰岛细胞分泌功能的影响,探讨石斛合剂防治糖尿病的机制。方法:分别用0%,5%,10%,20%的DC含药血清和13.3nmol.L-1的IGF-1(胰岛素样生长因子-1)孵育24h后,加入1.0mmol.L-1的STZ作用24h,采用放免法检测各组细胞的胰岛素分泌量及其对高糖刺激的反应性。结果:5%DC含药血清,对1.0mmol.L-1STZ损伤的胰岛细胞保护作用不明显,(P>0.05),10%,20%DC含药血清的保护作用显著,胰岛素分泌量较正常对照组明显增加(P<0.05),作用与IGF-1阳性对照组类似(P>0.05)。结论:10%,20%DC含药血清能保护胰岛细胞,减轻STZ对其所致的损伤。
OBJECTIVE: To investigate the effect of Dendrobium Mixture (DC) on the secretory function of pancreatic islet cells damaged by streptozotocin (STZ), and to explore the mechanism of Dendrobium Mixture in preventing and treating diabetes mellitus. Methods: After incubated with 0%, 5%, 10%, 20% DC serum and IGF-1 (insulin-like growth factor-1) at 13.3nmol.L-1 for 24 hours respectively, 1.0mmol.L-1 Of STZ 24h, using radioimmunoassay to detect the amount of insulin secretion of each group of cells and its response to high glucose stimulation. Results: The protective effect of 5% DC containing serum on islet cells damaged by 1.0mmol.L-1STZ was insignificant (P> 0.05). The protective effect of 10% and 20% DC containing serum was significant and the insulin secretion was more than normal The control group was significantly increased (P <0.05), similar to the IGF-1 positive control group (P> 0.05). Conclusion: 10%, 20% DC containing serum can protect islet cells and relieve the damage caused by STZ.
