目的研究肝细胞生长因子(HGF)及其受体c-Met在肝细胞癌门静脉癌栓中的表达,探讨HGF及c-Met的表达与门静脉癌栓形成与发展的关系。方法用半定量逆转录-聚合酶链反应(RT-PCR)技术检测HGF mRNA及c-Met mRNA在20例肝细胞癌门静脉癌栓及其癌旁组织、10例无门静脉癌栓形成的肝细胞癌和癌旁组织及10例正常肝组织中的表达。结果HGF及c-Met在肝癌及癌栓中的阳性表达率均显著高于正常肝组织及癌旁组织,差异有统计学意义(P<0.05),HGF及c-Met在癌栓中及肝癌中表达的阳性率比较差异无统计学意义(P>0.05),HGF与c-Met在各组织中的表达阳性率高低差异无统计学意义(P>0.05)。HGF及c-Met在肝癌及癌栓中的表达量均显著高于正常肝组织及癌旁组织,差异有统计学意义(P<0.05),HGF及c-Met在癌栓中表达量显著高于其在肝癌中表达量(P<0.05),HGF与c-Met在各组织中的表达量差异无统计学意义(P>0.05)。结论HGF及c-Met参与了肝癌的形成,而肝癌患者HGF及c-Met量的升高可能是肝癌发展形成门静脉癌栓的原因之一,HGF及c-Met可作为门静脉癌栓形成的监测指标。
Objective To investigate the expression of hepatocyte growth factor (HGF) and its receptor c-Met in hepatocellular carcinoma (HCC) tumor thrombus and to investigate the relationship between the expression of HGF and c-Met and the formation and development of portal vein tumor thrombus. Methods HGF mRNA and c-Met mRNA were detected by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) in 20 cases of hepatocellular carcinoma with portal vein tumor thrombus and its adjacent tissues and 10 cases of portal vein tumor thrombus Cancer and adjacent tissues and 10 cases of normal liver tissue expression. Results The positive rates of HGF and c-Met in HCC and tumor thrombus were significantly higher than those in normal liver tissues and paracancerous tissues (P <0.05). The positive rates of HGF and c-Met in HCC and HCC (P> 0.05). There was no significant difference in the positive expression rates of HGF and c-Met between the two groups (P> 0.05). The expression of HGF and c-Met in hepatocellular carcinoma and tumor thrombus were significantly higher than those in normal liver tissues and adjacent normal tissues (P <0.05). The expression of HGF and c-Met in tumor thrombus was significantly higher There was no significant difference in the expression of HGF and c-Met between the two groups (P> 0.05). Conclusions HGF and c-Met are involved in the formation of hepatocellular carcinoma. The increase of HGF and c-Met in patients with hepatocellular carcinoma may be one of the reasons for the development of portal vein tumor thrombus in hepatocellular carcinoma. HGF and c-Met can be used as the monitoring of portal vein tumor thrombus formation index.
