目的研究低剂量PCB77急性暴露对小鼠氧化还原状态的影响,从中获取后续研究的经验性指标。方法 60只雄性昆明小鼠随机分为5组,对照组(C)、0.036 mg/kg bw剂量组(Ⅰ)、0.18 mg/kg BW剂量组(Ⅱ)、0.9 mg/kg BW剂量组(Ⅲ)和4.5 mg/kg bw剂量组(Ⅳ),观察PCB77不同剂量和染毒1、3、7 d后对小鼠肝、脾组织中谷胱甘肽过氧化物酶(GSH-Px)活力、超氧化物岐化酶(SOD)活力和丙二醛(MDA)含量的影响。结果①与对照组相比,不同染毒时间各浓度组小鼠肝、脾组织GSH-Px、SOD的相对活性都低,除I浓度组外,差异均有统计学意义。且各浓度组不同时间肝、脾组织GSH-PX、SOD活性差异有统计学意义。②与对照组比较,PCB77Ⅰ-Ⅳ处理组小鼠肝脏组织中MDA含量上升,差异有统计学意义;各浓度组MDA含量随时间的增加而下降。结论低剂量的PCB77短期暴露可引起小鼠肝脏中GSH-PX及SOD活性、MDA含量的明显改变,且存在时间和剂量的依赖性。调节这些在氧化-抗氧化防御系统平衡中起重要作用指标的相关分子或通路或许是进一步研究这类物质毒性的重要目标。
Objective To study the effect of low dose PCB77 exposure on the redox state in mice and obtain the empirical indexes of subsequent studies. Methods Sixty male Kunming mice were randomly divided into five groups: control group (C), 0.036 mg / kg bw dose group (Ⅰ), 0.18 mg / kg BW dose group (Ⅱ) and 0.9 mg / ) And 4.5 mg / kg bw dose group (Ⅳ). The effects of different doses of PCB77 on the activity of glutathione peroxidase (GSH-Px) in mice liver and spleen after 1, The activity of oxidative superoxide dismutase (SOD) and the content of malondialdehyde (MDA). Results ① Compared with the control group, the relative activities of GSH-Px and SOD in the liver and spleen of mice in different concentration groups were all lower than those in the control group, with the exception of the I group. The concentrations of GSH-PX and SOD in liver and spleen of different concentration groups were significantly different at different time points. ② Compared with the control group, the content of MDA in the liver of PCB77Ⅰ-Ⅳ treatment group increased, the difference was statistically significant; MDA content in each concentration group decreased with time. Conclusion Short-term exposure of low-dose PCB77 can cause GSH-PX and SOD activity in liver and significantly change the content of MDA, and there is time-and dose-dependent. It may be an important goal to further study the toxicity of these substances by modulating these molecules or pathways that play an important role in the balance of the oxidation-antioxidant defense system.
