目的建立小鼠急性酒精性肝损伤模型并筛选敏感检测指标。方法实验分8个组,实验组小鼠采用一次经口灌胃50%的乙醇(12 ml/kg)后禁食4、6、8、10、12、16、20和24 h,相应对照组灌胃给予纯净水并禁食同等时间。血液生化指标检测包括血中三酰甘油(TG)、胆固醇(CHOL)、丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、碱性磷酸酶(AKP)、胆红素(TBIL)和Y-谷氨酰转酞酶(Y-GGT);16 h实验组和对照组血中高密度脂蛋白(HDL)和低密度脂蛋白(LDL)含量;并取动物肝脏,冰冻切片苏丹Ⅲ染色后,观察肝组织脂肪变性程度。结果 经口一次灌胃给予小鼠50%的乙醇后禁食16 h,实验组与对照组比较呈现明显的脂质代谢紊乱,包括血中TG含量显著升高、HDL值显著降低、病理肝组织脂肪变性程度明显加重。结论在该实验条件下,50%的乙醇经口灌胃一次后禁食16 h,可造成小鼠急性酒精性肝损伤模型,可通过测定血中TG、HDL含量及肝组织脂肪变性程度等指标进行模型评价。
Objective To establish a mouse model of acute alcoholic liver injury and screen sensitive detection indexes. Methods The experiment was divided into 8 groups. The mice in the experimental group were fasted for 4, 6, 8, 10, 12, 16, 20 and 24 h after oral administration of 50% ethanol (12 ml / Gavage with pure water and fasted for equal time.
