目的观察腹腔注射胰岛新生相关蛋白(INGAP)对链脲佐菌素(STZ)诱导的糖尿病(DM)小鼠的治疗作用。方法 C57BL/6小鼠腹腔注射STZ建立DM模型,将成模后小鼠随机分为INGAP组和磷酸盐缓冲液(PBS)对照组。INGAP组每日给予腹腔注射INGAP500μg;对照组注射等体积PBS。两组均隔日监测血糖至第34天,用实时荧光定量-PCR检测小鼠胰腺中葡萄糖转运蛋白-2(Glut-2)和胰腺十二指肠同源盒1(PDX-1)mRNA的表达。结果 INGAP组血糖未见明显下降;但与对照组比较,INGAP可以使DM小鼠胰腺中PDX-1、Glut-2mRNA的表达增加(P<0.05)。结论腹腔注射INGAP不能使STZ诱导的DM小鼠血糖下降,但能够使胰腺中的PDX-1、Glut-2表达量提高。
Objective To observe the therapeutic effect of intraperitoneal injection of islet neonatal related protein (INGAP) on streptozotocin (STZ) -induced diabetic (DM) mice. Methods C57BL / 6 mice were injected intraperitoneally with STZ to establish DM model. The mice were randomly divided into INGAP group and phosphate buffered saline (PBS) control group. In INGAP group, intraperitoneal injection of INGAP 500μg was given daily. In control group, equal volume of PBS was injected. Blood glucose was monitored every other day in both groups until day 34, and the expression of glucose transporter-2 (Glut-2) and pancreatic duodenal homeobox 1 (PDX-1) mRNA in the pancreas was detected by real-time fluorescence quantitative PCR . Results No significant decrease of blood glucose was observed in INGAP group. However, compared with control group, INGAP could increase the expression of PDX-1 and Glut-2 mRNA in DM mouse pancreas (P <0.05). Conclusion Intraperitoneal injection of INGAP can not decrease blood glucose in STZ-induced DM mice, but it can increase the expression of PDX-1 and Glut-2 in pancreas.
